Gene evolution has been thought to be chiefly impelled by gene duplication and rearrangement mechanisms since 1970s3. This study illustrates a new driven force for gene evolution: that is, head-on collision between DNA replication and transcription. Thus, it raises a question whether there are other important factors driving gene evolution. At the small end of the scale, the authors show the positive selection for a gene that is head-on oriented with replication, which breaks through the thought limited to the evolutionary selection for co-oriented genes.
On the other hand, the conclusion reached in this paper is only applied to Bacillus subtilis, leaving question that if it holds true for other prokaryotes and even eukaryotes. Take yeast as an example, transcription associated mutation is influenced by the orientation in study of Kim et al.4; while the collisions negatively affect DNA transcription independent of orientation in paper of Azvolinsky et al.5. These contradictory results demonstrate that systematic studies of other organisms are needed to decide whether the results of this paper can extend to other organisms.
